FFPE tissue sections can be processed through the Dewax and HIER Buffer, prior to manual staining or automated staining on the Epredia Autostainer. Formaldehyde fixation negatively impacts immunoreactivity of antigens/epitopes. While enzymatic digestion reverses this status for only a few antigens/epitopes, HIER has been established as the pre-treatment method of choice for many antigens/epitopes to improve their immunoreactivity with antibodies designed to detect their expression in FFPE tissues.
